High-throughput cloning platform in Yarrowia lipolytica.

Context

Yarrowia lipolytica is a model organism for metabolic studies of fatty acid biosynthesis and heterologous protein production. The development of high-throughput screening techniques will accelerate knowledge of this yeast, particularly regarding the annotation and function of numerous uncharacterized genes.

Objectives

The PLAYA project aims to leverage the knowledge acquired on Yarrowia lipolytica to establish a high-throughput cloning platform facilitating the screening of extensive mutant libraries. Concurrently, novel selection and screening methodologies will be developed to enhance the efficiency and scope of functional genomics in this yeast.

Expected results

Development of methods for identifying genomic regions of interest. Construction of chassis strains for library creation. Development of a high-throughput screening method for lipid secretion.
