3DLIPID

Tissue‑ and subcellular imaging by cryo‑TOF‑SIMS of oilseed germination, for 3D lipidomic mapping

Biomolécule

Context

Metabolomic approaches have advanced our understanding of numerous biological mechanisms by probing closely to the molecular phenotype. However, one key limitation of commonly used “bulk” approaches is their lack of spatial information—despite the fact that the biosynthesis and function of many metabolites depend precisely on cellular compartmentalization within different organelles and specialized membranes.

Objectives

The objective of this project is to establish a proof of concept for the technical feasibility of analyzing plant material using cryo-TOF-SIMS, thereby opening new analytical perspectives across various fields.

Expected results

TOF-SIMS (Time-of-Flight Secondary Ion Mass Spectrometry) has the potential to enable 3D visualization of lipid distribution at the subcellular level, thereby facilitating advancements in understanding the associated biological mechanisms by combining the advantages of microscopy and lipidomics.
